1. Describe the punch method. ELLAAA
In the process of filling hand-fill capsules, the pharmacist uses the punch method, which
entails repeatedly pounding the open end of the base of the capsule into the powder
while making sure that the level is half the size of the capsule body. Finally, the cap or
other half of the capsule will be reapplied to seal the capsule. The weight of each
capsule is then measured using an empty capsule, and powders are then added or
subtracted from the weight until the desired weight is achieved.
2. Give the different sizes of hard gelatin capsules and their approximate capacity by
capsule size. Give examples of diluents and disintegrants that are frequently added

Hard capsules come in a range of standard sizes, which are denoted by numerals
ranging from 000 to 5. The standard size capsules and their respective contents (in mg)
are as follows: 00 (950 mg), 00 (650 mg), 0 (450 mg), 1 (300 mg), 2 (250 mg), 3 (200
mg), 4 (150 mg), and 5. (100mg). Microcrystalline cellulose (MCC) is frequently utilized
in tablet and capsule formulations as a binder/diluent. Starch is an example of a classic
disintegrant, while croscarmellose sodium, crospovidone, and sodium starch glycolate
are examples of "super disintegrants."
3. Compare the procedures for compounding hard gelatin capsules with tablets and

When we compound hard gelatin capsules with tablets we use the punch method to produce the
capsules. The powder is placed on a clean surface like a sheet of paper or glass and is formed
and gathered together by a spatula. Taking the capsule you punch the tablet vertically to fill the
powder. When the capsule is finished we then take the cap and cover the top of the capsule and
then push until you hear a click sound. For gelatine capsules with liquid ingredients we’ll use a
micropipette so that an accurate measurement can be obtained for the capsule. We then placed
the capsule in a machine where the base and the cap are separated. The machine will then
completely mix the liquid which will eventually spread over the bases and fill the capsules
evenly.
