The bacteria present in water are retained on the membrane. The membrane with the bacteria so
isolated is then put in contact with a suitable nutrient (M-Endo’s medium) which inhibits the growth of

temperature of 37°C for a period of 20 hours. The bacteria of coliform group if present in water are
developed into visible colonies which can be counted with the help of microscope.

Serial dilution involves the process of taking a sample and diluting it through a series of standard volumes
of sterile diluent, which can either be distilled water or 0.9 % saline. Then, a small measured volume of

In attached growth processes, the microorganisms responsible for the conversion of organic material
or nutrients are attached to an inert packing material. The organic material and nutrients are removed
from the waste-water flowing past the attached growth also known as a biofilm. Packing materials used
in attached growth processes include rock, gravel, slag, sand, redwood, and a wide range of plastic
and other synthetic materials. Attached growth processes can also be operated as aerobic or
anaerobic processes. The packing can be submerged completely in liquid or not submerged, with air
or gas space above the biofilm liquid layer. The most common aerobic attached growth process used
is the trickling filter in which wastewater is distributed over the top area of a vessel containing non-
submerged packing material

Slow sand filtration is a type of centralized or semi-centralized water purification system. A well-
designed and properly maintained slow sand filter (SSF) effectively removes turbidity and pathogenic
organisms through various biological, physical and chemical processes in a single treatment step. Only
under the prevalence of a significantly high degree of turbidity or algae-contamination, pre-treatment
measures (e.g. sedimentation) become necessary. Slow sand filtration systems are characterized by a
high reliability and rather low lifecycle costs. Moreover, neither construction nor operation and
maintenance require more than basic skills. Hence, slow sand filtration is a promising filtration method
for small to medium-sized, rural communities with a fairly good quality of the initial surface water
source.

The objective of secondary treatment is the further treatment of the effluent from primary treatment to
remove the residual organics and suspended solids. In most cases, secondary treatment follows primary
treatment and involves the removal of biodegradable dissolved and colloidal organic matter using
aerobic biological treatment processes. Aerobic biological treatment (see Box) is performed in the
presence of oxygen by aerobic microorganisms (principally bacteria) that metabolize the organic matter
in the wastewater, thereby producing more microorganisms and inorganic end-products (principally
CO2, NH3, and H2O). Several aerobic biological processes are used for secondary treatment differing
primarily in the manner in which oxygen is supplied to the microorganisms and in the rate at which
organisms metabolize the organic matter.
High-rate biological processes are characterized by relatively small reactor volumes and high
concentrations of microorganisms compared with low rate processes. Consequently, the growth rate of
new organisms is much greater in high-rate systems because of the well controlled environment. The
microorganisms must be separated from the treated wastewater by sedimentation to produce clarified
secondary effluent. The sedimentation tanks used in secondary treatment, often referred to as
secondary clarifiers, operate in the same basic manner as the primary clarifiers described previously. The
