1 Overview
The Shimadzu SPD-40 and SPD-40V are high-performance, multi-function ultra-violet
visible spectrophotometric detectors, for use in high performance liquid chromatograph. 1
The SPD-40V, with a D2 lamp and W lamp as light source, provides highly sensitive
analysis throughout the entire wavelength range.
Each instrument has 3 measurement modes - single wavelength, dual wavelength and
wavelength scanning.
The dual wavelength mode performs simultaneous measurement of two wavelengths, and
can provide chromatograms of both wavelengths, or a chromatogram of one wavelength
and one ratio chromatogram. In wavelength scanning mode, the absorbance spectrum
can be measured while the flow is stopped.
1.1 Features
An improved optical system and a high-order digital filter have enhanced the instrument's
signal to noise ratio.
n Advanced functions
Mobile phases have higher absorbances in shorter wavelengths than in longer ones, which
allows variation in the ambient temperature to affect the absorbance of the mobile phase
in the cell and leads to change in the baseline or peak areas. The instrument stabilizes
analysis by controlling the temperature of the flow cell.
Replacement of the lamps does not require troublesome optical axis adjustment. The
instrument enables users to monitor the total lamp usage time and to see when to
replace the lamps with a glance.
By using the absorbance spectrum of the built-in holmium oxide filter and the emission
line spectrum of the D2 (deuterium) lamp, the instrument checks the wavelength accuracy
across a range from ultraviolet light to visible light, which provides easy performance
verification.

1 Overview
The built-in cell ID memory in the flow cell stores information on the type of the cell.
This enables the instrument to record the type of the cell used in an analysis in the
analysis data and provides better analysis traceability.
The lamps also have built-in ID memories and allow correct management of their
illumination time. The illumination time and ID numbers of the lamps used in an analysis
are automatically recorded in the analysis data, which enhances analysis traceability.

When turning off the main power switch on the back of the instrument after 2
changing the parameters via panel operation, be sure to turn OFF the power
with the power button at the front of the instrument, and turn off the main
power switch. Otherwise, some of the changed parameters may return to their
original values.

*1 The button cannot be used if the instrument is linked with the system controller SCL-40. If SCL-40
is connected, pressing the power button of SCL-40 turns off the power of the entire system.
If CBM-40 or CBM-40lite is used, pressing the power button of the pump turns off the power of
the entire system.
Note, however, that the following will apply in the case of a system with multiple pumps.
- When using CBM-40lite: The system power can be turned off using the power button of the
pump where CBM-40lite is built in.
- When using CBM-40: The system power can be turned off using the power button of the pump
with the smallest optical link number.

Use the keys on the front to operate or configure the instrument. When you touch the
key, the status LED lights up and accepts input.

3 Operation
3.1 Precautions
• The flow cell must be installed in the detector Fbefore turning the power switch ON.
If the power switch is turned on with no cell installed, home position will not be
detected and the wavelength will be set incorrectly.
If this occurs, turn the power switch OFF, install the cell and turn the power ON. The
cell may be removed after the initial screen is displayed.
• When highly sensitive analysis is necessary, take into account the time needed for the
baseline to stabilize and turn the lamp on ahead of time.
After it is turned on, the lamp requires roughly 1 hour to stabilize for optimal
performance.
Keep the front cover closed during analysis. In high-sensitivity analysis, opening or closing
the front cover will cause the baseline to fluctuate. The noise level may increase if the
front cover is open.
Power is supplied.

3.2 Turning Power ON/OFF
2 When the power is turned ON, the following sequence of events occurs:
If an alarm sounds and a [WAVE CHECK NO GOOD] message is displayed on the screen:

3 Check to be certain that no air bubbles have been introduced into the flow cell,
and that no mobile phase or sample which absorbs light in the vicinity of 254nm
or 656nm remains in the cell.
The wavelength calibration function [WAVE CALIB] and wavelength accuracy check
function [WAVE CHECK] measure the intensity of light transmitted through the
cell in the vicinity of the 656nm and 254nm
emission lines of the D2 (deuterium) lamp and mercury lamp. The instrument
operates based on these intensity values. Consequently, if a sample that absorbs
ultraviolet or visible light remains in the cell, or if
large air bubbles are introduced, the amount of transmitted light becomes
exceedingly small and the instrument will not operate correctly.
Operate using a flow cell that has been flushed or filled with a mobile phase
which does not absorb visible and UV light, or that has been purged of air or
nitrogen and dried thoroughly.
5 When the calibration is complete, the [WAVE CHECK] will be run automatically to
recheck the wavelength accuracy. If [WAVE CHECK NO GOOD] appears once again,
turn off the power of the instrument and contact the nearest Shimadzu
branch/sales office or agent.
If an alarm sounds and a [FATAL USER DATA] message is displayed on the screen:
Press [CE] to clear the alarm. When this message is displayed, the time program, along
with the [LAMBDA] (wavelength) and certain other parameters, will be initialized
(replaced with default values).

n Wavelength
Specify the wavelength to be measured. The wavelength2 can set only in the the
dual-wavelength measurement mode. The SPD-40V uses a D2 lamp and a W lamp as the
light source that provides a 190 to 1000 nm wavelength range absorption spectrum. To
measure only the ultraviolet region, turn on the D2 lamp only.
See below for information about the the single wavelength mode.

• The detector has two of signal output connectors: [RECORDER] connector and
[INTEGRATOR] (integrating data processor) connector.
A Chromatopac or variable range recorder can be connected to either of the
two connectors, but a Chromatopac should normally be connected to the
[INTEGRATOR] connector. A fixed range recorder must be connected to the
[RECORDER] connector so that its recording range can be adjusted using the
instrument's recorder range setting function. If a Chromatopac is connect to
the [RECORDER] connector, the Chromatopac's input full scale will be
equivalent to 100 times the absorbance value set for the recorder range.
Accordingly, the absorbance value set for the range should be about 1/80 of
the expected maximum peak absorbance.
• When the wavelength is changed:
The [RECORDER] connector signal output level changes to 0V for 4 seconds,
then returns to the previous signal level. This results in a mark at the point
of wavelength change on the chromatogram. However, since this could
interfere with area calculations, the [INTEGRATOR] connector output is
designed to maintain its signal level unchanged during the 4 second time
period.

3.3 Setting Parameters
n Response
How responsive the digital filter is influences the S/N and responsiveness of the
instrument.
Noise increases as the filter's response becomes faster, and decreases as it becomes slowly.
A quicker response of the filter results in a larger noise. A slower response value leads
to a smaller noise.

Setting "-" to polarity inverts the polarity of absorbance of recorder output terminal and
integrator output terminal from positive (negative) to negative (positive). Normally use
"+" for polarity. If the absorbance of the mobile phase is higher than the absorbance
of the component targeted for analysis (using the indirect absorbance detection method),
set the polarity to "-".

3.3 Setting Parameters
n Recycle valve
The instrument controls the temperature of the flow cell and thus mitigates the impact
of ambient temperature variation on the baseline. The temperature control is effective
when the mobile phase has a high absorbance or when the sample has a strong
temperature dependence.
If the temperature of the column is higher than the set temperature of the flow
cell, the heat of the mobile phase is transferred and the temperature of the
flow cell may not drop to the set temperature. In this case, increase the set
temperature of the flow cell.
If the flow cell temperature exceeds the upper limit of 50 ° C, turn off the cell
temperature control.
Ready Check
The ready check ensures that analysis starts after heat balance is established at
temperature-controlled locations.
The ready check is available for the flow cell and polychromator.

line specific to the lamp (universal physical constant). The traceability of universal

When the instrument is connected to the system controller SCL-40/CBM-40, the power
control of the instrument follows the system controller, and the shutdown and startup
functions operate from the system controller throughout the system.
If you turn off the power to the instrument individually with the setting [1], the
connection to the system controller will also be lost. Therefore, the system
startup function does not turn on the instrument. You need to turn the power
on again by pressing and holding the power button on the instrument. The
system shutdown function is still enabled with setting [1]. However, if the power
is turned off by the shutdown function, turning on the power by pressing the

• Files which do not contain scanned data cannot be plotted. If an empty file
is specified, the message [DATA NOT EXIST] is displayed. Similarly, plotting
cannot be performed if there is no data in the background file ([SCAN FILE]
= 0).
• If the scanning range includes 370 nm in the SPD-40/SPD-40V or 700 nm in the
SPD-40V, noise caused by insertion of the second-order diffraction filter will
appear at these wavelengths.
• Spectrum data cannot be output to the [INTEGRATOR] connector.
• Spectrum data is lost when the detector is turned off.
• Changing the scanning conditions ([SCAN BGN], [SCAN END], [SCAN STEP]) from
those that were used when scanning was started can prevent data from being
plotted correctly.
When the absorbance of the mobile phase varies substantially over the scanning
wavelength range, the effect of the instrument's wavelength reproducibility error (0.2 nm

Example
In the mobile phase absorbance spectrum shown below, the absorbance change per
wavelength is greatest in the section between 210 nm and 220 nm.
The rate of change per nanometer in this section is (1.0 - 0.5) /(220 - 210) = 0.05 AU/nm.
Therefore, the maximum spectrum error which could be introduced by wavelength
reproducibility error is 0.2 nm ´ 0.05 AU/nm = 0.01AU. To obtain a spectrum in which
this error may be ignored, the absorbance maximum of the peak of interest should be
about 100 times the possible error, or about 1 AU.

2 When connecting two or more circuits to the terminals, use the wires as shown
below.
The instrument provides one event cable. When more than 2 cables are required, use
the following cables.
Before using the cable, strip the jacket (1) at the end approximately 10 mm.
• Cable with stranded wire: 0.3 mm2 to 1.25 mm2 (AWG22 to 16), diameter of
single wire thicker than f 0.18.
• For a stranded cable, twist the ends well.
The cable with stranded wire is suitable to prevent disconnection.
Turn off the main power switch on the back of the instrument and unplug
the power cable prior to performing inspections and maintenance when
instructed.
Otherwise, fire, electric shock or malfunction may occur.
When replacing parts, use only the parts listed in "1.2 Component Parts" and
"5.2 Consumable Parts/Maintenance Parts/Replacement Parts".
If any other parts are used, injury or malfunction may occur.

damaged and become unusable.
Necessary Parts

3 While the mobile phase is flowing, look at the inside of the flow cell in the
direction of the arrow to check that no bubble or dirt is in the cell.
4 Insert the syringe needle (G) on the tip of the syringe (H) and turn it clockwise
to fix it in place.
5 Remove the tubing on the cell outlet tubing (with a coupling) end.
6 Connect the syringe (H) to the cell outlet tubing (C). (Attach the male nut
0.8MN-PEEK (F) to the syringe needle and connect the coupling 1.6-0.8C (E).)
When using Inert cell, UHPLC Inert cell or Low Dispersion Inert cell, connect the
coupling to the cell inlet tubing.
8 Flush the flow cell with distilled water or deionized water using a syringe to
purge the rinsing solution (2-propanol).

• Plastic tweezers (with no dirt on the end)
• Air blower (for the lens) (Part No.: 086-78801-02)
If plastic tweezers are not available, use some kind of alternative item such as
a toothpick for removing. In this case, use something soft so that it does not
scratch the lens or cell window.

If use of the gaskets is continued without replacing, fluid may begin to leak.
1 Loosen the male nut (A) on the column side, and remove the flow cell inlet tubing
(B).
Remove the flow cell outlet tubing (C) in the same way.
2 Remove the left and right setscrews (D) using a flathead screwdriver.
3 Remove the lens (E) and gasket (F) using plastic tweezers.
3 Put the removed lenses in 2-propanol and clean them with an ultrasonic bath
for 5 minutes.
4 Attach the cleaned lenses and two new cell gaskets to the cell.
1 Use a piece of wiping paper soaked in 2-propanol to remove dirt inside the cell
housing (G).
2 If dust is attached to the lenses or gaskets, remove it using an airblower (for the
lens) while securing the lenses or gaskets with plastic tweezers so as not to be
blown away by the air.
If an air blower (for the lens) is not available, use a piece of wiping paper
soaked in 2-propanol to remove the dust.
3 Attach the new gaskets and lenses in that order using plastic tweezers, and attach
the setscrews using a flathead screwdriver.
4 Connect the syringe adapter to the flow cell outlet tubing. (Attach the male nut

5 Attach the syringe (K) with mobile phase solution for analysis to the syringe
needle, and slowly push the plunger to check for a fluid leak.
If there is a fluid leak, rinse the lenses again, and then tighten the
setscrews again so that they are not loose.
6 Loosen the male nut (A), and remove the syringe needle.
Necessary tools

• Plastic tweezers (with no dirt on the end)
• Air blower (for the lens) (Part No.: 086-78801-02)
If plastic tweezers are not available, use some kind of alternative item such as a toothpick
for removing. In this case, use something soft so that it does not scratch the lens or cell
window.
Necessary parts
If use of the gaskets is continued without replacing, fluid may begin to leak.
1 Loosen the male nut (A) on the column side, and remove the flow cell inlet tubing
(B). Remove the flow cell outlet tubing (C) in the same way.
2 Remove the left and right setscrews (D) using a flathead screwdriver.
3 Remove the lens (E) and gasket (F) using plastic tweezers.
1 Use a piece of wiping paper soaked in 2-propanol to remove dirt inside the cell
housing (G).
2 If dust is attached to the lenses, cell window or new gaskets, remove it using an
air blower (for the lens) while securing the lenses or new gaskets with plastic
tweezers so as not to be blown away by the air.
If an air blower (for the lens) is not available, use a piece of wiping paper
soaked in 2-propanol to remove the dust.
3 Attach the new gaskets and lenses in that order using plastic tweezers, and attach
the setscrews using a flathead screwdriver.
4 Connect the syringe adapter to the flow cell inlet tubing (B). (Attach the male nut
0.8MN-PEEK (I) to the syringe needle(J) and connect the coupling 1.6-0.8C
(H).)Connect the flow cell outlet tubing (C) to the tubing to the drain.
When using Inert cell, UHPLC Inert cell or Low Dispersion Inert cell,

The guaranteed service life of the D2 lamp is 2000 hours of use or 1 year from
the date of purchase, whichever is the shorter.
• The guaranteed service life of the W lamp is 1200 hours of use or 1 year from 4
the date of purchase, whichever is the shorter.
• Note that the average lifetime of W lamps is 2000 hours and therefore there
are some lamps broken before 2000 hours of operation.
Allow the instrument to cool down adequately before performing the lamp
replacement procedure.
The lamp compartment is extremely hot just after turning off the lamp.
When replacing the lamp, be careful not to touch the glass portion of the
lamp directly with your fingers.
Wrap the lamp in cotton gauze when carrying it. If the lamp becomes dirty,
wipe it with an ethanol-moistened lens cleaner.
During replacement work, be sure to keep the surfaces of the mirror and filter
(applicable to only SPC-40V) clean.
A dirty surface of the mirror or filter prevents accurate detection.

Necessary Parts
2 Unscrew the two top cover installation screws (B), and remove the top cover (A).
3 Loosen the four screws (D) holding the radiating fin assembly in the lamp
compartment, and remove the fin assembly (C).
The D2 lamp (F), mirror (G), filter (H) (for SPD-40V only) and W lamp (I) (for SPD-40V
only) will now be visible inside the lamp housing (E).
1 Remove the connector (A) of the D2 lamp and then the lead wire (C) from the
clamp (B).
2 Remove the short screw (E) and long screw (F) fastening the D2 lamp and then
remove the D2 lamp.
• The nut on the long screw (G) has been set to a particular position. Do not
loosen or move it.
• Avoid dropping the screws into the detector chassis; it is difficult to retrieve
them. However, be sure to retrieve them if they are dropped.
• If the D2 lamp is tight and it is difficult to remove it, screw in screws in two
screw holes (D) gradually and evenly to remove the D2 lamp from the lamp
housing.
Allow the instrument to cool down adequately before performing the lamp
replacement procedure.
The lamp compartment is extremely hot just after turning off the lamp.
During replacement work, be sure to keep the surfaces of the mirror clean.
A dirty surface of the mirror prevents accurate detection.
3 Fit the new D2 lamp (H) in place, and secure it with the screws.
When positioning the lamp, hold it with gauze to keep the surfaces clean. If
the lamp gets dirty, accurate analysis will not be possible.
Accurate readings cannot be taken when the glass surface is dirty.
Wipe off fingerprints on the glass using a cloth moistened with alcohol.
Be careful not to touch the glass parts directly with your hands when replacing
the lamp.
Wrap the lamp in cotton gauze when carrying it. When the lamp is dirty, wipe
it off with lens paper soaked in ethanol.
4 Put the lead wire (C) in the notch (I) and then fix it with the clamp (B) before
connecting the connector (A) back.
Ensure that the cable (C) is fitted into the notch (I) in the lamp housing, with
the 3-pin connector (A) located outside the housing.
If the cable is not in the notch, it could be broken, resulting in shorting or
lamp failure. (The same applies for the W lamp.)
Be sure to fix the lead wire (C) with the clamp (B).
Failure to do so may cause the lead wire to be broken and result in
malfunction.
5 Replace the fin assembly and top cover of the lamp housing and detector
respectively, and tighten the screws.
Attach the top cover in the correct orientation by aligning it with the hollow
of the instrument. (Tighten it with screws.)
Incorrect attachment may cause ultraviolet light to escape.
1 Insert the power plug to the outlet and turn ON the main power switch on the
back.
The initial screen will be displayed in a while.
4 Press [CE].
The initial screen is displayed.
1 Disconnect the 2-pin connector (A) at the end of the W lamp cable.
2 Loosen the three lamp screws (C), and remove the W lamp (B).
When removing the screws, do not drop the screws into the detector chassis. If
you drop it, be sure to remove it.
Allow the instrument to cool down adequately before performing the lamp
replacement procedure.
The lamp compartment is extremely hot just after turning off the lamp.

3 Place the new lamp in position in the lamp socket, matching the projection (E)
on the socket to the notch (D) in the lamp flange.
Be careful not to touch the glass parts directly with your hands when replacing

Wrap the lamp in cotton gauze when carrying it. When the lamp is dirty, wipe
it off with lens paper soaked in ethanol.
5 Put the lead wire (F) in the notch (G) and connect the 2P connector (A) back.
Ensure that the cable is fitted into the notch (G) in the lamp housing, with
the 2-pin connector (A) located outside the housing.
If the cable is not in the notch, it could be broken, resulting in shorting or
lamp failure. (The same applies for the D2 lamp.)
6 Replace the fin assembly and top cover of the lamp housing and detector
respectively, and tighten the screws.
Attach the top cover in the correct orientation by aligning it with the hollow
of the instrument. (Tighten it with screws.)

4 Press [Enter].
The numeric keypad is displayed.

To display the [PARAMETER] setting group,
press [→] three times in the initial screen. [LAMBDA 1] is displayed.
10 Press [Enter].

The screen displays the voltages of the
reference light and sample light.
[REF: If the [REF: ***mV] value is at
least "500mV", your are finished.
If the value is lower than "500mV", go on to step 14.
14 Increase the [W LAMP POWER] value in the order of [1], [2], and [3].
To display [W LAMP POWER], press [→] three times in the initial screen and then press
[↓] several times.
Increase the [W LAMP POWER] value until the [REF: ***mV] value exceeds "500mV".
You are finished when the value exceeds "500mV".
Necessary parts
2 Make sure there are no air bubbles in the flow cell, and then install the cell into
the detector.
7 Press [Enter].
Wavelength accuracy calibration is run automatically.

4 If the setting displayed in the upper right is not "1", press [Enter], [1], and then
[Enter].
The setting displayed in the upper right changes to "1" (D2 lamp ON).
9 Remove the standard flow cell, attach the filter holder (A) to the detector and
place the cover (B) over the filter holder. Then, press [Enter].
12 Press [Enter].
[Input Filter Abs] is displayed.
13 Input the absorbance given on the certificate for the inputted wavelength and
then press [Enter].
15 Remove the absorbance calibration filter from the filter holder, and attach the
filter holder cover.
19 Press [CE] to return to the initial screen and then press [ZERO] followed by [Enter]
to check that the displayed absorbance changes to "0".
20 Insert the absorbance calibration filter (C), then attach the cover.

21 Record the absorbance value shown on the initial screen.
22 Confirm that the error between the displayed absorbance and the absorbance
value given on the certificate of the absorbance calibration filter is within ±3 %.
This finishes the calibration of the absorbance accuracy.
If the initial screen is not displayed, press [CE] to display the initial screen.
2 Press [↓] once, move the cursor to [VP] and press [→].
3 Press [↓] several times, move the cursor to [CALIBRATION] and press [→].
When wiping off moisture, do not press the leak sensor. When a sensor unit
touches a wall, the sensor sensitivity is lowered.
9 From the value read in step 7, calculate the threshold value of the leak sensor.
How to obtain the leak sensor threshold:
(Maximum value-100) ´ 0.7 + 100
In the example screen here, [142] is calculated.
1 Dip a piece of cloth in a dilute neutral detergent and twist firmly to remove
excess liquid. Use this cloth to scrub the soiled area of the exterior surface of

the instrument.
2 Dip a piece of cloth into water and twist firmly to remove excess liquid. Use
this cloth to wipe away all the remaining detergent. Use a dry cloth to remove
all moisture from the exterior surface of the instrument.
Do NOT allow spilled water to remain on the instrument surface, and do not
use alcohol or thinner-type solvents to clean the surfaces.
These can cause rusting and discoloration.
*1 Noise level in dual wavelength mode varies depending on the wavelength. For example, use
sensitivity 0.005 AUFS max. at 250 nm. Generally, for wavelengths with small optical outputs, noise
level increases accordingly.
