1 Overview
The Shimadzu SPD-40 and SPD-40V are high-performance, multi-function ultra-violet
visible spectrophotometric detectors, for use in high performance liquid chromatograph. 1
The SPD-40V, with a D2 lamp and W lamp as light source, provides highly sensitive
analysis throughout the entire wavelength range.
Each instrument has 3 measurement modes - single wavelength, dual wavelength and
wavelength scanning.
The dual wavelength mode performs simultaneous measurement of two wavelengths, and
can provide chromatograms of both wavelengths, or a chromatogram of one wavelength
and one ratio chromatogram. In wavelength scanning mode, the absorbance spectrum
can be measured while the flow is stopped.

1.1 Features

n Excellent S/N ratio performance

An improved optical system and a high-order digital filter have enhanced the instrument's
signal to noise ratio.

n Advanced functions

The dual wavelength mode simultaneously provides chromatograms of two wavelengths


or ratio chromatograms, and the wavelength scanning mode provides absorbance spectra.
There is also a full set of Time Program functions.

n Improved stability in analysis through flow cell temperature control

Mobile phases have higher absorbances in shorter wavelengths than in longer ones, which
allows variation in the ambient temperature to affect the absorbance of the mobile phase
in the cell and leads to change in the baseline or peak areas. The instrument stabilizes
analysis by controlling the temperature of the flow cell.

n Easy replacement and inspection (maintenance) of consumables

Replacement of the lamps does not require troublesome optical axis adjustment. The
instrument enables users to monitor the total lamp usage time and to see when to
replace the lamps with a glance.
By using the absorbance spectrum of the built-in holmium oxide filter and the emission
line spectrum of the D2 (deuterium) lamp, the instrument checks the wavelength accuracy
across a range from ultraviolet light to visible light, which provides easy performance
verification.

1 Overview

n Cell ID and lamp ID functions

The built-in cell ID memory in the flow cell stores information on the type of the cell.
This enables the instrument to record the type of the cell used in an analysis in the
analysis data and provides better analysis traceability.
The lamps also have built-in ID memories and allow correct management of their
illumination time. The illumination time and ID numbers of the lamps used in an analysis
are automatically recorded in the analysis data, which enhances analysis traceability.

2 Parts Identification and Function
2.1 Front

2.5 Names and Functions of Display and Operation Area

2.5 Names and Functions of Display and Operation Area


This instrument is controlled through the keypad of the operation area. The display area
allows verification of the instrument status.
The operation area has two input modes: [Arrow key input mode] to enable screen
transfer, and [Numerical key input mode] to enable value input.
Pressing [Enter] switches between these input modes.

When turning off the main power switch on the back of the instrument after 2
changing the parameters via panel operation, be sure to turn OFF the power
with the power button at the front of the instrument, and turn off the main
power switch. Otherwise, some of the changed parameters may return to their
original values.

confirmation screen as shown in the figure. Holding it down

with the smallest optical link number.

3 Operation
3.1 Precautions

n Precautions before Operation

• The flow cell must be installed in the detector Fbefore turning the power switch ON.
If the power switch is turned on with no cell installed, home position will not be
detected and the wavelength will be set incorrectly.
If this occurs, turn the power switch OFF, install the cell and turn the power ON. The
cell may be removed after the initial screen is displayed.
• When highly sensitive analysis is necessary, take into account the time needed for the
baseline to stabilize and turn the lamp on ahead of time.
After it is turned on, the lamp requires roughly 1 hour to stabilize for optimal
performance.

n Precautions during Operation

Keep the front cover closed during analysis. In high-sensitivity analysis, opening or closing
the front cover will cause the baseline to fluctuate. The noise level may increase if the
front cover is open.

n Precautions after Operation

To prevent clogging of the flow cell:


Dusty or clogged flow cells are the most frequent cause of detector problems. After
analyzing a highly concentrated sample, flush the flow cell thoroughly, using plenty of
mobile phase. When a buffer solution is used as the mobile phase, wash the flow cell
with water after completing analysis. Buffer solutions crystallize upon evaporation, and
can clog the flow cell and tubing.

3.2 Turning Power ON/OFF

3 Check to be certain that no air bubbles have been introduced into the flow cell,
and that no mobile phase or sample which absorbs light in the vicinity of 254nm
or 656nm remains in the cell.
The wavelength calibration function [WAVE CALIB] and wavelength accuracy check
function [WAVE CHECK] measure the intensity of light transmitted through the
cell in the vicinity of the 656nm and 254nm
emission lines of the D2 (deuterium) lamp and mercury lamp. The instrument
operates based on these intensity values. Consequently, if a sample that absorbs
ultraviolet or visible light remains in the cell, or if
large air bubbles are introduced, the amount of transmitted light becomes
exceedingly small and the instrument will not operate correctly.
Operate using a flow cell that has been flushed or filled with a mobile phase
which does not absorb visible and UV light, or that has been purged of air or
nitrogen and dried thoroughly.

4 Execute wavelength calibration, referring to [WAVE CALIB] in Section 5.6, VP


Functions.

5 When the calibration is complete, the [WAVE CHECK] will be run automatically to
recheck the wavelength accuracy. If [WAVE CHECK NO GOOD] appears once again,
turn off the power of the instrument and contact the nearest Shimadzu

If an alarm sounds and a [FATAL USER DATA] message is displayed on the screen:
Press [CE] to clear the alarm. When this message is displayed, the time program, along
with the [LAMBDA] (wavelength) and certain other parameters, will be initialized
(replaced with default values).

n Wavelength

Specify the wavelength to be measured. The wavelength2 can set only in the the
dual-wavelength measurement mode. The SPD-40V uses a D2 lamp and a W lamp as the
light source that provides a 190 to 1000 nm wavelength range absorption spectrum. To
measure only the ultraviolet region, turn on the D2 lamp only.

• The detector has two of signal output connectors: [RECORDER] connector and
[INTEGRATOR] (integrating data processor) connector.
A Chromatopac or variable range recorder can be connected to either of the
two connectors, but a Chromatopac should normally be connected to the
[INTEGRATOR] connector. A fixed range recorder must be connected to the
[RECORDER] connector so that its recording range can be adjusted using the
instrument's recorder range setting function. If a Chromatopac is connect to
the [RECORDER] connector, the Chromatopac's input full scale will be
equivalent to 100 times the absorbance value set for the recorder range.
Accordingly, the absorbance value set for the range should be about 1/80 of
the expected maximum peak absorbance.
• When the wavelength is changed:
The [RECORDER] connector signal output level changes to 0V for 4 seconds,
then returns to the previous signal level. This results in a mark at the point
of wavelength change on the chromatogram. However, since this could
interfere with area calculations, the [INTEGRATOR] connector output is
designed to maintain its signal level unchanged during the 4 second time
period.

How responsive the digital filter is influences the S/N and responsiveness of the
instrument.
Noise increases as the filter's response becomes faster, and decreases as it becomes slowly.
A quicker response of the filter results in a larger noise. A slower response value leads
to a smaller noise.

n Controlled Cell Temperature

The instrument controls the temperature of the flow cell and thus mitigates the impact
of ambient temperature variation on the baseline. The temperature control is effective
when the mobile phase has a high absorbance or when the sample has a strong
temperature dependence.

If the temperature of the column is higher than the set temperature of the flow
cell, the heat of the mobile phase is transferred and the temperature of the
flow cell may not drop to the set temperature. In this case, increase the set
temperature of the flow cell.
If the flow cell temperature exceeds the upper limit of 50 ° C, turn off the cell
temperature control.

• To deactivate the temperature control: Enter "0" in the temperature setting.

Ready Check
The ready check ensures that analysis starts after heat balance is established at
temperature-controlled locations.
The ready check is available for the flow cell and polychromator.

emission line of the D2 lamp at 656.1 nm.

on the instrument (holmium filter).

When the wavelength is confirmed to be inaccurate, [WAVE CHECK NOGOOD]

How to measure the wavelength accuracy


SPD-40/SPD-40V use zero-order light (0 nm) and the emission line (656.1 nm) of a
deuterium lamp (D2 lamp) for calibrating the wavelength. The wavelength accuracy can
be secured by using those wavelengths as the reference for adjustments.
The technique to calibrate wavelengths using the emission line of a deuterium lamp (D2
lamp) is stipulated in the JIS standard as follows.

Measure the light intensity of a low-pressure mercury lamp or a deuterium lamp to

In addition to being controlled by the system controller, it is also possible to

turn off the power individually from the instrument.

When the instrument is connected to the system controller SCL-40/CBM-40, the power
control of the instrument follows the system controller, and the shutdown and startup
functions operate from the system controller throughout the system.

If you turn off the power to the instrument individually with the setting [1], the
connection to the system controller will also be lost. Therefore, the system
startup function does not turn on the instrument. You need to turn the power
on again by pressing and holding the power button on the instrument. The
system shutdown function is still enabled with setting [1]. However, if the power
is turned off by the shutdown function, turning on the power by pressing the
power button of the instrument is disabled, and the system must be started up
from the system controller.

If the [OPERATION MODE] is set to [1] and connected as SPD-20A/SPD-20AV, the


power control function by the system controller is not available. Therefore, you
can always operate the power button of the instrument regardless of the setting
of this function.

check the wavelength accuracy at 241 nm
during start-up. If a mobile phase with
strong absorption near 241 nm is used, the
wavelength accuracy check can not be
performed correctly. In that case, the wavelength check of 241 nm can be skipped by
setting it to "OFF".
The screen displays the current setting in the upper right. The setting "0" does not check
the wavelength accuracy at 241 nm while the setting "1" checks the wavelength accuracy
at 241 nm.
Pressing [Enter] displays the numeric keypad. Input the desired value and press [Enter].

n Change the setting when performing the wavelength accuracy test with manual
wavelength change 《Z WAVE》

Normally, the absorbance baseline is set to connect smoothly even if the wavelength is
changed, but it is necessary to change the setting when performing the wavelength

The current setting is displayed in the upper right. Normally, use "0", and even if the
wavelength is changed, the absorbance baseline will be connected smoothly. When
changed to "1", the baseline fluctuates according to the change in light intensity when

3.8 Measuring in Single Wavelength Mode

n When a Chromatopac (Integrator) is connected to the [RECORDER] Connector

• Connect the Chromatopac (integrator) to the [RECORDER] connector.


• Peaks approximately 100 times the range can be recorded. Set the range at about 1/80
of the expected maximum peak absorbance during measurement. Normally, the
recorder range should be set to about 0.005 to 0.04 AUFS, and the full scale plot
adjusted using the [ATTEN] attenuation control of the Chromatopac.

• The relationship in terms of plot full scale between the range and the Chromatopac's
[ATTEN] setting (when the Chromatopac is connected to the [RECORDER] connector) is
as follows:

Plot full scale absorbance = Range ´ 2ATTEN/10

Example: If range = 0.01 AUFS and ATTEN = 2

Plot full scale absorbance = 0.01 AUFS ´ 22/10 = 0.004 AUFS

For simultaneous processing of two channels by a Chromatopac:


In the dual-wavelength measurement mode, chromatograms for each wavelength can be
recorded simultaneously. The wavelength linked to the [INTEGRATOR] connector (the
shorter wavelength end) is regarded as channel 1 and that linked to the [RECORDER]
connector (the longer wavelength end) as channel 2. By using the settings displayed in
the table at right for [RANGE] setting value and [AUX RANGE], the signal to output
voltage ratio is equalized for both channels.

Note that [RESPONSE] has no effect on peak area. Peak area does not change

Assuming no baseline drift, [RT THR] should be set at a value equal to 1-5% of the full
scale measuring range (the Range Value). If the baseline drifts, for example in gradient
analysis, execute an auto zero operation in the time program shortly before the peak of
interest elutes, and set [RT THR] to a value larger than the expected drift.

Be sure to actuate the auto zero function at the beginning of each analysis.
Absorbance values used in the calculation of ratios are relative to the auto zero
point.

represent the change in absorbance, relative to the auto zero point, in wavelengths l1

Check the absorbance on the initial screen and stop feeding the liquid when the
specimen reaches the cell.
• When using a syringe:
After stopping feeding of the liquid, perform injection.

• If the scanning range includes 370 nm in the SPD-40/SPD-40V or 700 nm in the
SPD-40V, noise caused by insertion of the second-order diffraction filter will
appear at these wavelengths.
• Spectrum data cannot be output to the [INTEGRATOR] connector.
• Spectrum data is lost when the detector is turned off.
• Changing the scanning conditions ([SCAN BGN], [SCAN END], [SCAN STEP]) from
those that were used when scanning was started can prevent data from being
plotted correctly.

n Peak Intensity Required for Spectrum Scanning

When the absorbance of the mobile phase varies substantially over the scanning
wavelength range, the effect of the instrument's wavelength reproducibility error (0.2 nm

Max) on the spectrum data can be significant. To minimize this effect, the absorbance
value of the peak of interest should be as high as possible. More specifically, absorbance
at the maximum absorbance wavelength of the peak of interest should be about 100
times the maximum absorbance change of the mobile phase per 0.2 nm increment of
the scanning wavelength range.

Example
In the mobile phase absorbance spectrum shown below, the absorbance change per
wavelength is greatest in the section between 210 nm and 220 nm.

The rate of change per nanometer in this section is (1.0 - 0.5) /(220 - 210) = 0.05 AU/nm.
Therefore, the maximum spectrum error which could be introduced by wavelength
reproducibility error is 0.2 nm ´ 0.05 AU/nm = 0.01AU. To obtain a spectrum in which
this error may be ignored, the absorbance maximum of the peak of interest should be
about 100 times the possible error, or about 1 AU.

2 When connecting two or more circuits to the terminals, use the wires as shown

The instrument provides one event cable. When more than 2 cables are required, use
the following cables.
Before using the cable, strip the jacket (1) at the end approximately 10 mm.

• Cable with single wire: f 0.4 tof 1.2 (AWG26 to 16)

• Cable with stranded wire: 0.3 mm2 to 1.25 mm2 (AWG22 to 16), diameter of
single wire thicker than f 0.18.
• For a stranded cable, twist the ends well.
The cable with stranded wire is suitable to prevent disconnection.

4 Maintenance
4.1 Periodic Inspection and Maintenance
It is necessary to perform periodic inspections of this instrument to ensure its safe use.
It is possible to have these periodic inspections performed by Shimadzu service
representatives on a contractual basis.
For information regarding the maintenance inspection contract, contact your Shimadzu
representative.

Turn off the main power switch on the back of the instrument and unplug
the power cable prior to performing inspections and maintenance when
instructed.
Otherwise, fire, electric shock or malfunction may occur.

Never remove the main cover.


Otherwise, injury or malfunction may occur. Contact your Shimadzu
representative to remove the main cover.

When replacing parts, use only the parts listed in "1.2 Component Parts" and
"5.2 Consumable Parts/Maintenance Parts/Replacement Parts".
If any other parts are used, injury or malfunction may occur.

4.1.1 Prior to Inspection and Maintenance

• Replace the mobile phase in the flow lines with water.


• Wipe away any dirt from the front panel and the main cover.
• Wipe away any dirt from the keypad with tissue paper or a soft cloth moistened with
water.

Be sure to turn off the power of the equipment when plugging or unplugging
the flow cell cable connector.

4.2.1 Removing the Flow Cell

1 Open the front cover.

2 Remove the coupling from the coupling holder (B).

1 Turn up the connector side and then install the flow cell to the instrument.

2 While holding the flow cell fitted to the installation surface, tighten the two cell
mounting screws (C) alternately.

3 Insert the connector (B) into the instrument. With the coupling holder (A), fix
the coupling to the instrument.

Make sure that the equipment is turned off before plugging the connector.

4 Connect the inlet tubing of the flow cell directly to the column.
Via the coupling, connect the outlet tubing of the flow cell to the drain tubing.

When performing the above operation, do not let any air enter the flow lines.

5 Close the front cover.

If the baseline shows a large noise or large drift, check the flow cell. If the check finds
some bubbles or dirt, try the simple cleaning.
After cleaning the flow cell with the syringe according to the following procedure, if the
light intensity is not improved, replace the column with the coupling and pump a rinsing
solution (2-propanol) into the flow cell assembly for 2 hours.

Pump a solution from the inlet tubing into the flow cell.
If pumped from the outlet tubing to the inlet tubing, the flow cell may get
damaged and become unusable.

4.3 Flow Cell Inspection/Cleaning

4 Insert the syringe needle (G) on the tip of the syringe (H) and turn it clockwise
to fix it in place.

5 Remove the tubing on the cell outlet tubing (with a coupling) end.

6 Connect the syringe (H) to the cell outlet tubing (C). (Attach the male nut
0.8MN-PEEK (F) to the syringe needle and connect the coupling 1.6-0.8C (E).)

When using Inert cell, UHPLC Inert cell or Low Dispersion Inert cell, connect the
coupling to the cell inlet tubing.

7 Gently push in the syringe's plunger (I).


The liquid will be injected into the cell interior and clean the cell.

If an aqueous solvent (especially a solvent containing buffer salts) is used as


the mobile phase, purge the solvent in the cell with water before flushing
with 2-propanol.
If an organic solvent is mixed with salts, crystals will form and clog the tubing.

8 Flush the flow cell with distilled water or deionized water using a syringe to
purge the rinsing solution (2-propanol).

9 Perform step 3 to check for bubbles and dirt.

4.4 Disassembling and Cleaning the Flow Cell


If dirt and air bubbles cannot be removed by performing a simple cleaning, disassemble
and clean the flow cell. Also, disassembling and cleaning the flow cell is recommended
every 2 years.

Necessary tools
• Flathead screwdriver (Part No.: 086-10504-18)
• Plastic tweezers (with no dirt on the end)
• Air blower (for the lens) (Part No.: 086-78801-02)

If plastic tweezers are not available, use some kind of alternative item such as
a toothpick for removing. In this case, use something soft so that it does not
scratch the lens or cell window.

the setscrews using a flathead screwdriver.

4 Connect the syringe adapter to the flow cell outlet tubing. (Attach the male nut
0.8MN-PEEK (I) to the syringe needle(J) and connect the coupling 1.6-0.8C 4
(H).)Connect the flow cell inlet tubing to the tubing to the drain.

When using Inert cell, UHPLC Inert cell or Low Dispersion Inert cell,
use the supplied coupling 1.6-0.8C PEEK. In this case, connect the
coupling to the cell inlet tubing.

5 Attach the syringe (K) with mobile phase solution for analysis to the syringe
needle, and slowly push the plunger to check for a fluid leak.

If there is a fluid leak, rinse the lenses again, and then tighten the
setscrews again so that they are not loose.

6 Loosen the male nut (A), and remove the syringe needle.

It is necessary to replace the lens or cell window if dirt on the lens or cell window cannot
be removed after disassembling and cleaning the flow cell. The gaskets should be
replaced at the same time. Also, it is recommended to replace the lenses or cell window
every 2 years.

Before storing the flow cell, pump methanol into it. Then, drain the methanol with the
syringe and dry the inside of the flow cell or fill the flow cell with methanol and seal
the cell before storing it.

The SPD-40 has a D2 lamp, and the SPD-40V has a D2 lamp and a tungsten lamp. The
lamps need to be replaced periodically.
As the D2 lamp and W lamp in this detector approach the end of their life, their intensity
decreases and baseline noise increases. Replace the lamps with new ones using the
guaranteed service life as a guide.

n Guaranteed Service Life of the Lamp

Guaranteed service life of the D2 lamp : 2000 hours

The guaranteed service life of the D2 lamp is 2000 hours of use or 1 year from
the date of purchase, whichever is the shorter.

Guaranteed service life of the W lamp : 1200 hours


Average lifetime : 2000 hours

• The guaranteed service life of the W lamp is 1200 hours of use or 1 year from 4
the date of purchase, whichever is the shorter.
• Note that the average lifetime of W lamps is 2000 hours and therefore there
are some lamps broken before 2000 hours of operation.

Before replacing a lamp, turn the detector power switch OFF.


Failing to do so may result in fire, electric shock, or malfunction.

Allow the instrument to cool down adequately before performing the lamp
replacement procedure.
The lamp compartment is extremely hot just after turning off the lamp.

lamp directly with your fingers.
Wrap the lamp in cotton gauze when carrying it. If the lamp becomes dirty,
wipe it with an ethanol-moistened lens cleaner.

Be careful not to break the lamp.


Do not impart shocks or vibrations to the lamp.

Be sure to install the lamp cover before switching on the lamp.


The lamp cannot be switched on while the lamp cover is removed.
If the lamp cover is removed without switching off the lamp, the lamp is
extinguished automatically.

During replacement work, be sure to keep the surfaces of the mirror and filter
(applicable to only SPC-40V) clean.
A dirty surface of the mirror or filter prevents accurate detection.

4.7.1 Removal of Top Cover and Radiating Fin Assembly

1 Turn OFF the main power switch on the back.

2 Unscrew the two top cover installation screws (B), and remove the top cover (A).

The fcover assembly's two screws cannot be removed.

3 Loosen the four screws (D) holding the radiating fin assembly in the lamp
compartment, and remove the fin assembly (C).
The D2 lamp (F), mirror (G), filter (H) (for SPD-40V only) and W lamp (I) (for SPD-40V
only) will now be visible inside the lamp housing (E).

The fin assembly's four screws cannot be removed.

4.7.2 Deuterium Lamp Replacement

1 Remove the connector (A) of the D2 lamp and then the lead wire (C) from the
clamp (B).

2 Remove the short screw (E) and long screw (F) fastening the D2 lamp and then
remove the D2 lamp.

• The nut on the long screw (G) has been set to a particular position. Do not
loosen or move it.
• Avoid dropping the screws into the detector chassis; it is difficult to retrieve
them. However, be sure to retrieve them if they are dropped.
• If the D2 lamp is tight and it is difficult to remove it, screw in screws in two
screw holes (D) gradually and evenly to remove the D2 lamp from the lamp
housing.

Before replacing a lamp, turn the detector power switch OFF.


Failing to do so may result in fire, electric shock, or malfunction.

Allow the instrument to cool down adequately before performing the lamp
replacement procedure.
The lamp compartment is extremely hot just after turning off the lamp.

Be careful not to break the lamp.

3 Fit the new D2 lamp (H) in place, and secure it with the screws.

When positioning the lamp, hold it with gauze to keep the surfaces clean. If
the lamp gets dirty, accurate analysis will not be possible.
Accurate readings cannot be taken when the glass surface is dirty.
Wipe off fingerprints on the glass using a cloth moistened with alcohol.

Be careful not to touch the glass parts directly with your hands when replacing
the lamp.
Wrap the lamp in cotton gauze when carrying it. When the lamp is dirty, wipe
it off with lens paper soaked in ethanol.

4.7 Replacing the Lamp

4 Put the lead wire (C) in the notch (I) and then fix it with the clamp (B) before
connecting the connector (A) back.

Ensure that the cable (C) is fitted into the notch (I) in the lamp housing, with
the 3-pin connector (A) located outside the housing.
If the cable is not in the notch, it could be broken, resulting in shorting or
lamp failure. (The same applies for the W lamp.)

Be sure to fix the lead wire (C) with the clamp (B).
Failure to do so may cause the lead wire to be broken and result in
malfunction.

5 Replace the fin assembly and top cover of the lamp housing and detector
respectively, and tighten the screws.

Securely tighten the screw of the radiation fin.


A loose screw may worsen the performance.

Attach the top cover in the correct orientation by aligning it with the hollow
of the instrument. (Tighten it with screws.)
Incorrect attachment may cause ultraviolet light to escape.

it off with lens paper soaked in ethanol.

4 Re-tighten the three lamp screws alternately.


The W lamps are fixed.

4.8 Wavelength Accuracy Calibration


Measure the peak wavelengths in the emission line spectra produced by the deuterium
lamp, and calibrate them to minimize wavelength variance.

1 Enclose either of water, methanol, or acetonitrile in the cell.


Use the pump to pump water, methanol, or acetonitrile through the flow cell, or fill the
cell with any of these three using a syringe and syringe adapter.
Refer to "4.3 Flow Cell Inspection/Cleaning" P.105 for the proper use of the syringe and
syringe adapter.

2 Make sure there are no air bubbles in the flow cell, and then install the cell into
the detector.

4.9 Absorbance Accuracy Calibration


Calibrate absorbance accuracy so as to minimize the difference between the absorbance
value displayed on the instrument when the tested absorbance calibration filter is in
place and the absorbance value of the filter.

9 Remove the standard flow cell, attach the filter holder (A) to the detector and
place the cover (B) over the filter holder. Then, press [Enter].

10 The display shows [Input Filter WL].

11 After about 5 seconds, when [Insert


Filter] and [Input Filter Abs] are
displayed, remove the cover and then
attach the absorbance calibration filter

13 Input the absorbance given on the certificate for the inputted wavelength and

15 Remove the absorbance calibration filter from the filter holder, and attach the
filter holder cover.

16 Press [CE] to return to the initial screen.

17 Display the [PARAMETER] setting group of the auxiliary function (FUNCTION)


group.
To display the [PARAMETER] setting group, press [→] three times in the initial screen.
[LAMBDA 1] is displayed.

18 Press the [Enter] key to display the


numeric keypad. After entering the
wavelength used for calibration, press
the [Enter] key.

to check that the displayed absorbance changes to "0".

20 Insert the absorbance calibration filter (C), then attach the cover.

22 Confirm that the error between the displayed absorbance and the absorbance
value given on the certificate of the absorbance calibration filter is within ±3 %.
This finishes the calibration of the absorbance accuracy.

4.10 Leak Sensor Calibration


Re-calibration is required if the result of [LEAK SENSOR CHECK] fails the component
validation.

The leak sensor calibration must be performed by the system administrator.


The leak sensor detects leaked mobile phase when the tubing causes the leakage.
When the output value from the sensor exceeds the level set at "threshold
value", an error occurs and temperature control stops.

1 Press [→] on the initial screen.

If the initial screen is not displayed, press [CE] to display the initial screen.

2 Press [↓] once, move the cursor to [VP] and press [→].

3 Press [↓] several times, move the cursor to [CALIBRATION] and press [→].

4 Input the password.

5 Press [↓] several times until [LEAK


THOLD] is displayed.

6 Soak the sensor unit (A) at the bottom


of the leak sensor in water.
The leak sensor value (Actual Level)
increases by detecting water.

7 Read the maximum value of the leak


sensor value (Actual Level).
Example: Here, the maximum value is
[160].

8 Wipe off the moisture around the leak


sensor.
The leak sensor value (Actual Level)
decreases.

When wiping off moisture, do not press the leak sensor. When a sensor unit
touches a wall, the sensor sensitivity is lowered.

4.11 Exterior Cleaning

9 From the value read in step 7, calculate the threshold value of the leak sensor.
How to obtain the leak sensor threshold:

4.11 Exterior Cleaning


If the instrument cover or front panel becomes dirty, wipe it clean with a soft dry cloth
or tissue paper.
For persistent stains, clean the exterior using the following procedure.

1 Dip a piece of cloth in a dilute neutral detergent and twist firmly to remove
excess liquid. Use this cloth to scrub the soiled area of the exterior surface of

the instrument.

2 Dip a piece of cloth into water and twist firmly to remove excess liquid. Use
this cloth to wipe away all the remaining detergent. Use a dry cloth to remove
all moisture from the exterior surface of the instrument.

Do NOT allow spilled water to remain on the instrument surface, and do not
use alcohol or thinner-type solvents to clean the surfaces.
These can cause rusting and discoloration.
