GRAM Staining :

1. Staphylococcus Aurues
 Blood Agar
 Circular, convex, opaque, smooth surface, white in colour
 Non motile
 Gram +ve coccus
 Catalase +ve, Coagulase +ve
2. Bacillus Subtilis
 Nutrient Agar
 Dry, flat, irregular, with lobate margin, white in colour
 Gram +ve bacillus
 Motile
 Catalase +ve
3. E. Coli
 MacConkey Agar
 Circular, moist, smooth with entire margin, pink, non mucoid, pink
 Gram –ve bacillus
 Motile
 Catalase +ve
 Ferment all sugar except sucrose
 Indole +ve , Citrate –ve, Urease –ve
4. Klebsiella sp.
 MacConkey Agar
 Circular, moist, smooth with entire margin, pink to red, mucoid
 Gram –ve bacillus
 Non motile
 Ferment all sugars
 Catalase +ve
 Indole -ve , Citrate +ve, Urease +ve
5. Pseudomas sp.
 MacConkey Agar
 Pale or colourless
 Gram –ve bacillus
 Earthy smell
 Motile
 Ferments glucose
 Motile
 Catalase +ve
 Oxidase +ve
6. Proteus
 MacConkey Agar
 Smooth, pale or colourless
 Fishy smell
 Gram –ve bacillus
 Motile
 Ferment only glucose
 Catalase +ve
 Indole +ve

 Isolation and differentiation of urinary organisms like E.Coli, Klebsiella sp.
5. Thiosulphate Citrate Bile Salt Agar/ TCBS Agar
 Dark green
 Used for selective isolation of vibrio cholerae
6. Sabouraud Dextroxe Agar/ SDA
 Slight yellow
 Used for isolation of dermatophytes
