In this study we examined the plasma levels of Aβ, proNGF, tPA, neuroserpin, metallo-proteases and inflammatory molecules in individuals with Down syndrome, with and without dementia. We described associations between biomarkers and cognitive decline

Nerve growth factor metabolic dysfunction in Alzheimer's disease and Down syndrome
Trends in Pharmacological Sciences
This review summarizes the recent evidence supporting a CNS dysfunction of NGF's extracellular metabolism in Alzheimer's disease and Down syndrome. It also discusses the implications of such trophic disconnection on the phenotype of basal forebrain cholinergic neurons.

Intracellular Abeta pathology and early cognitive impairments in a transgenic rat model overexpressing human amyloid precursor protein: a multidimensional study
Acta Neuropathologica Communications
This study investigates the early development of the Aβ pathology and its associated cognitive impairments in a novel transgenic rat overexpressing a human APP transgene. Using high-magnification and super-resolution microscopy this investigation further contributes to resolve the controversy that Aβ is found intraneuronally.

Nerve growth factor metabolic dysfunction in Down's syndrome brains
Brain
Individuals with Down’s syndrome are at increased risk of developing Alzheimer’s disease in adulthood. Using post-mortem human brain tissue, fetal cortex cultures and Ts65Dn trisomic mice, in this study we revealed marked alterations in nerve growth factor metabolism in Down’s syndrome, consistent with similar defects previously reported in the Alzheimer’s disease brain.

This article describes evidence suggesting that a pro-inflammatory process and, consequently, the deregulation NGF extracellular metabolism could be amongst the earliest pathological events in the progression to AD.
