LABORATORY EXERCISE 3

BREAKING DORMACY OF APPLE SEEDS

I. Introduction

Seed dormancy allows seeds to overcome periods that are unfavorable for seedling established and is
therefore important for plant ecology and agriculture. Several processes are known to be involved in the
induction of dormancy and in the switch from the dormant to the germinating state.

There are two types of seed dormancy in general: seed coat (physical) dormancy and internal dormancy.
In seed coat dormancy, the seed coat prevents oxygen and/or water permeating into the seed. Sometimes,
dormancy is caused by inhibiting chemicals inside the seed. Seeds with seed coat dormancy can remain on/in
the ground without germinating until the seed coat allows water and oxygen to enter the seed or eliminate the

II. Objective/s

At the end of the exercise the student must be able determine and describe the best way to break
dormancy of apple seed; And understand the seed dormancy mechanism.

III. Materials

✓ 4-5 pcs of Fuji apple


✓ Lukewarm water
✓ 2 Transparent plastic containers (any shape medium size w/ flat bottom)
✓ Tissue/paper towels
✓ Hydrogen peroxide or Agua Oxinada (10/20 volume at least 100 ml)
✓ Water

IV. Procedure

1. Extract the seeds of the apple from the fruits. Cut the flesh on each side until the seeds on the center of the
fruit exposed. Do not cut the fruit in the center. You should able to extract at least 20 seeds
2. After all the seeds have extracted, wash them thoroughly with lukewarm water to remove the sugary
residues in the seed coat. Air dried the seeds after washing to remove the excess moisture.
3. Divide the seeds into two group (10-20 seeds per group). Label each plastic container with T1 and T2.
4. Follow the following methods for each seed group:

✓ T1: Soak the seeds in water for 2-4 hrs then remove the seed coat/testa of the seed. After soaking put the
seeds in a transparent container with moisten tissue/paper towel at the bottom then cover.

✓ T2: Soak the seeds in the hydrogen peroxide for 30 mins. After soaking put the seeds in a transparent
container with moisten tissue/paper towel at the bottom then cover.

5. Always check your set-up, do not let the tissue or paper towels to dry. Moisten with water when dried.
6. Place you set-up in a secure place in your home.
7. Photo document all your activities in the Exercise. This will be submitted in our GCR assigned as classwork.
8. Worksheet will be posted in our GCR to discuss the results of your Exercise.

9. Data to be gathered. Gather the following to be reported and discuss in the worksheet:

a. Date of set-up. Record the date when you applied the treatment for T1 & T2
b. Date of germination. Record the date when the 50% of the seed germinated for T1 & T2.
c. Percent germination. This will gather 14 days from set up using the formula:
% germination = (no. of seeds germinate / no. of seeds sown) x 100
