Primary and Secondary Screening - Crowded Plate

Primary screening helps determine microorganisms and organic acids in cells, while secondary screening provides quantitative and qualitative information about cells or desired strains. The crowded plate technique is used for primary screening to identify microorganisms capable of producing antibiotics. It involves plating a suitable aliquot of serial dilutions to yield 300-400 colonies on an agar plate, called a crowded plate. While simple and fairly rapid, it still requires further analysis and may not indicate antibiotic production against a specific organism. The technique was applied in a study that developed a novel method for quantifying soil suppression of fungi using a modified crowded plate approach to obtain antagonistic microorganisms.

Types of Screening
Primary screening helps to
determine the microorganisms
and organic acid in cells and
other desired organisms.

Secondary screening can be

used for determining the

desired organisms or strains.
Crowded Plate Techniques

This technique of primary screening helps

to determine the microorganisms that are
capable of producing the antibiotics or
not.

A “suitable aliquot” of “serial dilution” is

chosen that can produce 300 to 400
colonies when it is plated on the agar
plate, which is also called a crowded
plate.
