This document describes a method to determine the amount of dissolved oxygen in a water sample using Winkler's method. The method involves titrating a water sample against a standardized sodium thiosulfate solution and calculating the dissolved oxygen concentration based on the titration results. Key steps include standardizing the sodium thiosulfate solution and then titrating a water sample against it to determine the dissolved oxygen content.

DETERMINATION OF DISSOLVED OXYGEN IN WATER BY

WINKLER’SMETHOD

AIM:

To determine the amount of dissolved oxygen in the given water sample by

Winkler’s method. A standard solution of Potassium dichromate of strength 0.01N is given.

Titration – 1 Titration- 2
Burette Solution Sodium thiosulphate Standardised Sodium
(unknown) thiosulphate solution
Pipette Solution 20 ml of 0.01N of Potassium 100 ml Water sample
dichromate
Additional Reagent 5 ml of dilsulphuric 2ml of MnSO4 and 2ml of
acid and 10 ml KI solution alkali iodide and 2 ml of
Con H2SO4
Indicator Few drops of Starch solution Few drops of Starch solution
Endpoint Colour change from blue to Colour change from blue to
pale green pale green
Equivalent weight of oxygen= 8
Required Apparatus 300ml BOD bottle, 50 ml Burette, 20 ml
pipette, 250 ml Conical Flask, Funnel.
Titration I – Standardisation of Sodium Thiosulphate
Potassium Dichromate (Vs) Sodium thiosulphate Indicator : Starch.

Sl. Volume of Potassium Burette Readings (ml) Volume of Sodium Concordant value
No. dichromate (ml) Initial Final thiosulphate (ml) (ml)

1 20 0

2 20 0

Volume of Potassiumdichromate (V1) = 20 ml

Strength of Potassium dichromate (N1) = 0.01 N
Volume of Sodium thiosulphate (V2) =…….ml
Strength of Sodium thiosulphate (N2) = (V1N1/V2) = (20 x 0.01) / V2= ------- N

Titration II – Estimation of dissolved oxygen.

Water sample (Vs)Sodium thiosulphate Indicator :Starch

Sl. Volume of water Burette Readings (ml) Volume of Sodium Concordant value
No. sample (ml) Initial Final thiosulphate (ml) (ml)

1 20 0

2 20 0

Volume of Sodium thiosulphate (V1) = ------ml

Strength of Sodium thiosulphate (N1) = ------N
Volume of Water sample (V2) = 100 ml
Strength of Water sample (N2) = Strength of DO in water (N2) = (V1N1) / V2 = V1N1 / 100

Amount of dissolved oxygen in one litre of sample water = N2 x 8 x 1000 mg/L

Result:
Amount of dissolved oxygen in a given water sample = ---------------mg/L.
