4. Clean instruments (scissors, hemostats, tubing
stripper)
5. Plasma extractor

Procedure of Calibrating centrifuge
For preparation of PRP :
Platelet count on anticoagulated specimen

• No. of platelet in whole blood= platelets/µL X1000 X ml of whole blood.

• Prepare PRP at selected speed and time ( 2000 x g, 3 min + dec. time).

• Separate RBC and seal ( packed RBC+ 2 connected bags of empty & PRP).

• Strip the tubing several times so that they contain a representative sample of PRP.

Continued:
Seal off a segment on the tail. Then perform platelet count in the sealed
6 segement of PRP.
• No. of platelet in PRP= platelets/µL X1000 X ml of PRP.

• Calculate and record the % of yield= (no. of platelet in PRP X100)/no. of plt in WB.

• Repeat 3-4 X (different speed, time and donor) compare yields.

• Select the best. Record centrifuge ID, calibration settings, the date and name.

Preparation of Platelets ( Hard spin)
Centrifuge PRP, according to….,(5000 x g for 7 min + dec. time)

• Remove clamp. Express plasma leaving 55 to 60 ml platelet volume.

• Seal tubing with long tube attached to platelet bag. Let platelets rest for 1 hour.

• Place the platelets on an agitator for at least 1 hour.

• Strip the tubing several times so that they contain a representative sample ofplatelet
5 concentrate.

• Record no. of platelets in conc.= platelets/µL x1000x ml of platelets.

• Calculate and record the percentage of yield.

• Repeat with different donor PRP, speed and time, compare yields.

• Select the best. Record centrifuge ID, calibration settings, the date and name.

Notes:
• Quality control of centrifuge includes calibration of :
• speed and timer quarterly.
• temp check daily.
• Temp verification monthly.
• Calibrate each centrifuge individually.
When determining appropriate time and speed for centrifuge, consider
other products e.g:
Hematocrit and volume of packed RBCs.
Volume of plasma.
