Corynebacterium diphtheriae is a gram-positive, non-motile bacillus that causes diphtheria. It can be identified by its club-shaped morphology and presence of metachromatic granules stained dark purple with methylene blue. C. diphtheriae ferments glucose and maltose producing acid and is catalase and oxidase positive. It produces a powerful exotoxin that inhibits protein synthesis and kills cells. The exotoxin can be converted to a toxoid vaccine. C. diphtheriae infects the throat, nose and skin, causing a thick gray membrane.
