B. Test Tube #2-1% Hgci - Sir Forgot To Bring The Solution.: A. Salts of Heavy Metals

The document summarizes several protein tests using albumin solutions. Heavy metal salts like mercury chloride and lead nitrate cause precipitation that increases with excess reagent. Alkaloidal reagents like picric acid and tannic acid cause dissolution of precipitation in test tubes 1-4. Concentrated acids like hydrochloric, sulfuric, and nitric acid do not form precipitation. Heller's test with nitric acid and albumin forms a faint reaction and precipitation at the top. Adding 70% alcohol to albumin, gelatin, and peptone solutions does not cause a reaction.

Exercise #8

Proteins II

A. Salts of Heavy metals

1. Place 2 mL neutral 1% albumin solution in each of 3 test tubes.

a. [Link]
Test#1-
tube
1%#2-
(CH31% HgCI
COO)2Pb2

-sir forgot to bring the solution.

c. Test tube #3- 1% Cd(NO3)2

For both letter A and C the amount of

precipitation increases by an excess reagent.
B. Alkaloidal Reagents

1. Prepare 4 test tubes containing 2 mL each of 1% (sir told to use 0.2% because 1% it’s not available)
albumin solution. Add to the following reagents to the corresponding test tube using a drop at a time and
shaking after addition:

a. #1- saturated picric acid solution b. #2- 10% tannic acid solution

c. #3-10% TCA d. #4- 20% tungstic acid solution

The amount of ppt is dissolve in 1-4 test tubes

C. Concentrated Inorganic Acids

1. Prepare 3 test tubes containing 1mL each of 1% filtered albumin solution.

a. Test tube #1- concentrated HCI b. Test tube #2- concentrated H 2SO4

c. Test tube #3- concentrated HNO3

There is no formed of ppt in 1-3 test tubes.

D. Heller’s Test

1. Place 1 mL of 0.5% albumin solution in a test tube. Incline the test tube and add carefully 1 mL conc
HNO3.

a. Not shaken. b. 1 mL conc HNO 3

c. 0.5% albumin with distilled water

It gives faintly perceptible reaction to both

letter B and C.

And for the letter A there is a formed ppt at

the top of solution.

E. Alcohol
1. Place 1 mL of the following test solutions in separate test tubes:

a. 1% albumin b. 1% Gelatin c. 1% Peptone

Added 5 mL of 70% alcohol to each test tubes.
